Basically, will having uploaded your script to the black list site discourage potential managers/agents from signing you to shop that script around? Because it's already sort of "out there?" Or do they not care?

Can't see a negative. Even repped writers, like EdFury, have found success on The Black List.
Besides, even though it's on The Black List you can still benefit from an agent/manager/lawyer to package/negotiate the sale.

Besides, what do you care what a rep that doesn't rep you thinks?? Your goal is to promote your material in the most effective way possible. Utilizing all tools accessible to you.
If you've got good scores on The Black List, it just might motivate said rep to sign you quicker. Who knows. Take you out of the 'rep' market before someone else snatches you up.
And if said rep wants you to remove it from The Black List I would ask them 'why' and if the answer is in alignment with your career strategy, then, take it down.

There's not a manager on earth that doesn't understand using every avenue available to get your script into the hands of someone who could make it.
