I don't recall getting any specific guidelines when I had to write one, other than a suggestion that brevity would be appreciated.
As far as I know, the letters don't really come into play as far as picking the fellows. Maybe if the committee really is deadlocked they'd use the letters to choose a fellow, but my feeling was that it was, first and foremost, about the scripts.
In any case, it's a bit premature to worry about that. If you've made the semis (they haven't yet picked finalists, have they?), savor that. You'll certainly get a lot of attention if you make it to the next level, but you have every reason to be proud of a script that has survived this far in such a prestigious contest.
